Indications

  • Relief of signs & symptoms of osteoarthritis

Pharmacology

  • Consists of an immediate release Esomeprazole Magnesium layer
  • Enteric-coated Naproxen core
  • Naproxen is a NSAID with analgesic & antipyretic properties
  • Esomeprazole is a proton pump inhibitor

Dosage

  • Use lowest effective dose
  • Adult dose: 375 mg naproxen/20 mg of esomeprazole or 500 mg naproxen/20 mg of esomeprazole
  • Juvenile dose: 375 mg naproxen/20 mg of esomeprazole for >50 kg or 500 mg naproxen/20 mg of esomeprazole for >50 kg

Administration

  • Take at least 30 minutes before meals

Interaction

  • Concomitant use of NSAIDs may reduce the antihypertensive effect of ACE inhibitors, diuretics & beta-blockers
  • Concomitant use of this tablet and warfarin may result in an increased risk of a bleeding complication
  • Inhibits gastric acid secretion & may interfere with the absorption of certain drugs

Contraindications

  • Known hypersensitivity to any component of the tablet
  • History of asthmay urticaria or other allergic-type reactions after taking aspirin or other NSAIDs
  • Use during the peri-operative period in the setting of coronary artery bypass graft (CABG) surgery

Side Effects

  • Decreased occurrence of gastric ulcers and upper gastrointestinal adverse events compared to Naproxen alone
  • Possible small increased risk of arterial thrombotic events
  • Gastrointestinal side effects

Pregnancy & Lactation

  • Contraindicated during the third trimester of pregnancy
  • Not recommended during breastfeeding
  • May impair female fertility

Precautions & Warnings

  • Avoid combination with other NSAIDs
  • Minimize undesirable effects with lowest effective dose
  • Increased risk of GI bleeding in older people

Use in Special Populations

  • Use caution in elderly patients
  • Not recommended in patients with moderate to severe or severe renal impairment
  • Monitor patients with hepatic insufficiency closely
