Indications

Description
- Oral soluble film designed to be applied on top of the tongue and dissolve within 20 seconds
- Does not require water to aid dissolution or swallowing
- Contains the active ingredient Osedan base, a selective blocking agent of the serotonin 5-HT3 receptor type
Pharmacology
- Potent and highly selective 5HT3 receptor antagonist
- Blocks initiation of vomiting reflex by activating vagal afferents via 5HT3 receptors
- Antagonism of 5HT3 receptors on neurons in peripheral and central nervous system
Dosage
- Different dosage options for adults, pediatric patients, and geriatric patients
- Oral soluble film dosages for prevention of nausea and vomiting associated with highly and moderately emetogenic cancer chemotherapy
- Injection dosages for chemotherapy-induced nausea and vomiting, radiotherapy-induced nausea and vomiting, and postoperative nausea and vomiting
Administration
- Instructions for administration of oral soluble film
- Includes steps for tearing the pouch, placing the film on the tongue, and swallowing after it dissolves
Interaction
- Metabolized by hepatic cytochrome P-450 drug-metabolizing enzymes
- Inducers or inhibitors of these enzymes may change the clearance and half-life of Osedan
Contraindications
- Contraindicated in patients known to have hypersensitivity to the drug or any of its components
- Concomitant use of apomorphine
Side Effects
- Frequently reported adverse events include headache, constipation, and diarrhea
- Rare cases of anaphylaxis, bronchospasm, tachycardia, angina, hypokalemia, and shortness of breath reported
Pregnancy & Lactation
- Carcinogenic effects not seen in 2-year studies in rats and mice
- Caution advised when administered to nursing women due to excretion in breast milk of rats
Precautions & Warnings
- Hypersensitivity reactions reported in patients who have exhibited hypersensitivity to other selective 5-HT3 receptor antagonists
- Not a drug that stimulates gastric or intestinal peristalsis
Use in Special Populations
- Dosage adjustment same as for the general population in patients with impaired renal function
- Recommendation for a single maximal daily dose of 8 mg in patients with severe hepatic impairment
