Indications
- Reduction of symptoms of intermittent claudication
- Increased walking distance
Pharmacology
- Cilostazol is a preparation of quinolinone derivative
- Inhibits cellular phosphodiesterase III (PDE III)
- Suppresses cAMP degradation
- Increase in cAMP in platelets and blood vessels
- Inhibition of platelet aggregation
- Vasodilation
Dosage & Administration
- Recommended dosage: 100 mg bid
- Timing: at least half an hour before or two hours after breakfast and dinner
- Consideration of lower dose during coadministration with specific drugs
- Pediatric use not established
Interaction
- Omeprazole and Erythromycin increase systemic exposure
- Higher concentrations with concurrent use of Diltiazem
Contraindications
- Contraindicated in patients with congestive heart failure
- Contraindicated in patients with known or suspected hypersensitivity
Side Effects
- Common: headache, diarrhoea, vomiting, leg cramps, rash
- Less frequent: anorexia, edema
Pregnancy & Lactation
- No adequate and well controlled studies in pregnant women
- Transfer into milk reported in experimental animals
- Decision should be made to discontinue nursing or Cilostazol
Precautions & Warnings
- Use with caution in patients with any degree of heart failure
- No information on the concurrent use with Clopidogrel
