Indications
- Symptomatic treatment of chronic constipation in adults
Pharmacology
- Selective stimulator of 5-HT4 receptors
- No interaction with hERG channel or 5-HT1 receptors
- Stimulates motility by interacting with 5-HT4 receptors
Dosage & Administration
- Adults: 2 mg once daily
- Older people: Start with 1 mg once daily
- Children: Not recommended for use
Interaction
- Low interaction potential with CYP-mediated metabolism
- Weak substrate for P-glycoprotein
- May interact with potent inhibitors of P-gp
Contraindications
- Hypersensitivity to the active substance or excipients
- Renal impairment requiring dialysis
Side Effects
- Most frequently reported adverse reactions: headache, abdominal pain, nausea, diarrhea
- Majority of adverse events were mild to moderate in intensity
Pregnancy & Lactation
- Not recommended during pregnancy
- Effective contraception advised for women of childbearing potential
- Not recommended during breastfeeding based on animal studies
Precautions & Warnings
- Dose adjustment in severe renal impairment
- Caution in severe hepatic impairment
- Use of additional contraceptive method in case of severe diarrhea
- Not recommended for patients with rare hereditary problems of galactose intolerance
Use in Special Populations
- Patients with severe renal impairment: 1 mg once daily
- Patients with severe hepatic impairment: 1-2 mg once daily
Overdose Effects
- Symptoms may include headache, nausea, diarrhea

- Supportive measures may be required
