Indications

• Upper respiratory tract infections
• Lower respiratory tract infections
• Genito-urinary tract infections
• Skin and soft tissue infections
• Bone and joint infections

Composition

• 375 mg tablet: Amoxicillin 250 mg, Clavulanic Acid 125 mg
• 625 mg tablet: Amoxicillin 500 mg, Clavulanic Acid 125 mg
• 1 gm tablet: Amoxicillin 875 mg, Clavulanic Acid 125 mg
• Powder for Suspension: Amoxicillin 125 mg, Clavulanic Acid 31.25 mg
• Powder for Suspension (Forte): Amoxicillin 400 mg, Clavulanic Acid 57.5 mg
• 1.2 gm Injection: Amoxicillin 1 gm, Clavulanate Potassium 200 mg
• 0.6 gm injection: Amoxicillin 500 mg, Clavulanate Potassium 100 mg

Pharmacology

• Broad spectrum of bactericidal activity
• Inactivation of beta-lactamase enzymes
• Optimized absorption at the start of a meal
• Low levels of serum binding

Dosage

• Adults: 625 mg Tablet every 12 hours or 375 mg Tablet every 8 hours for usual dose
• Children: 25 mg/kg/day in divided doses every 8 hours
• Increased dosage for more severe infections

Administration

• Oral administration without regard to meals
• Intravenous administration over 2 minutes or slow intravenous infusion over 30 minutes
• Use within 20 minutes and discard any unused material

Interaction

• Prolongation of bleeding time and prothrombin time
• Reduced efficacy of oral contraceptives
• Potential interaction with allopurinol

Contraindications

• History of Penicillin hypersensitivity
• Cross-sensitivity with other beta-lactam antibiotics

Side Effects

• Diarrhoea, pseudomembranous colitis, indigestion, nausea, vomiting
• Hepatitis, cholestatic jaundice, urticarial and erythematous rashes

Pregnancy & Lactation

• No teratogenic effect in animal studies
• Limited use recommended in pregnancy, trace quantities in breast milk

Precautions & Warnings

• Caution in patients on anticoagulation therapy or with severe hepatic dysfunction
• Dosage adjustment in patients with renal impairment

Use in Special Populations

• Dose adjustment for patients with renal impairment
• Hepatic impairment monitoring

Overdose Effects

• Gastrointestinal symptoms and disturbance of fluid and electrolyte balances

Reconstitution

• IV reconstitution with Water for Injection BP
• Avoid mixing with certain solutions

Storage Conditions

• Store below 25°C, protected from light and moisture
• Refrigerate reconstituted suspension
• Use reconstituted vial within 20 minutes
