Description

  • Etacept is indicated for reducing signs and symptoms, inducing major clinical response, inhibiting the progression of structural damage, and improving physical function in patients with moderately to severely active rheumatoid arthritis (RA), psoriatic arthritis (PsA), ankylosing spondylitis (AS), juvenile idiopathic arthritis (JIA), and plaque psoriasis (PsO). It is a tumor necrosis factor (TNF) blocker and is administered as subcutaneous injections.

Dosage Instructions

  • The recommended dosage for adults with rheumatoid arthritis, psoriatic arthritis, and ankylosing spondylitis is 50 mg Etanercept per week administered either once weekly or 25 mg twice weekly as a subcutaneous injection. For plaque psoriasis, the dose is 50 mg once weekly or 25 mg twice weekly. Pediatric patients have different dosages based on their weight and age.

Administration

  • Etanercept should be administered as subcutaneous injections in the thigh, abdomen, or upper arm. Each new injection should be given at least 3 cm from a previous site. The injection should be performed under the supervision of a qualified healthcare professional.

Contraindications

  • Etanercept should not be initiated in patients with hypersensitivity to etanercept or to any component of the formulation, serious active infections, including chronic or localized infection, sepsis, or risk of sepsis.

Side Effects

  • Common side effects include infections, injection site reactions, allergic reactions, fever, itching, and autoantibody formation. Uncommon side effects include serious infections, low blood platelet count, skin cancer, angioedema, hives, eye inflammation, psoriasis, lung inflammation, and inflammation of blood vessels affecting multiple organs.

Pregnancy & Lactation

  • The safe use of etanercept during pregnancy and lactation has not been established. Use during pregnancy or lactation should be based on a clear need and a careful risk-benefit assessment.

Precautions & Warnings

  • Special precautions should be taken for allergic reactions, infections especially in patients with diabetes, tuberculosis, hepatitis-B, hepatitis-C, blood disorders, nervous system and eye disorders, congestive heart failure, cancer, chickenpox, latex allergy, alcohol abuse, Wegener's granulomatosis, and use with anti-diabetic medicine.

Overdose Effects

  • The maximum tolerated dose of Etacept has not been established in humans. Clinical trials have not observed dose-limiting toxicities, and there is no known antidote to Etacept.

  • Etacept should be stored in a refrigerator at 2-8°C, should not be frozen or shaken, and should be kept away from light and out of reach of children.
