Pharmacology
- In vitro activity against Gram positive aerobic bacteria
- Activity against some Gram positive anaerobic bacteria
- Activity against certain Gram negative bacteria
- Selective inhibition of bacterial protein synthesis
- Binding to the 23S ribosomal RNA of the 50S subunit of the bacterial ribosome
- Prevention of the formation of a functional 70S initiation complex
- Bacteriostatic against enterococci and staphylococci
- Bactericidal for the majority of streptococci strains
Dosage & Administration
- Oral administration with or without food
- IV infusion over a period of 30 to 120 minutes
- Recommended daily dosage for different infections
- No dose adjustment necessary when switching from IV to oral administration
- Inspection of IV infusion visually prior to administration
- No dose adjustment recommended for patients with mild-to-moderate hepatic impairment
Drug Interaction
- Reversible, nonselective inhibitor of monoamine oxidase
- Potential for interaction with adrenergic and serotonergic agents
- Enhancement of pressor response to certain agents
- Avoidance of large quantities of foods or beverages with high tyramine content
Contraindications
- Known hypersensitivity to Linezolid or any product components
- Concomitant use with monoamine oxidase inhibitors
- Uncontrolled hypertension
- Pheochromocytoma
- Thyrotoxicosis
- Carcinoid syndrome
- Use of directly and indirectly acting sympathomimetic agents
- Use of vasopressive agents
- Use of dopaminergic agents
- Use of serotonin re-uptake inhibitors
- Use of tricyclic antidepressants
Side Effects
- Most adverse events were mild to moderate
- Common adverse events include diarrhea, headache, and nausea
- Other adverse events include hypertension, dyspepsia, abdominal pain, pruritus, and tongue discoloration
Pregnancy & Lactation
- Use during pregnancy if potential benefit justifies potential risk to the fetus
- Presence in breast milk
- Consideration of developmental and health benefits of breastfeeding
Precautions & Warnings
- Monitoring of complete blood counts
- Ophthalmic evaluation for symptoms of visual impairment
- Careful medical history for Clostridium difficile-associated Diarrhea
- Monitoring for potential increases in blood pressure
- Immediate medical evaluation for specific symptoms
- Dose adjustment if hypoglycemia occurs
