Indications

Pharmacology

Proton pump inhibitor
Suppression of gastric acid secretion
Inhibition of H+/K+-ATPase in gastric parietal cell
Increased acid control compared to racemic proton pump inhibitors
Enteric-coated pellet formulation of esomeprazole magnesium
Absorption within approximately 1.5 hours after oral administration
Approximately 90% systemic bioavailability with once daily dosing
Extensive liver metabolism by CYP enzyme system
97% plasma protein binding
Plasma elimination half-life of approximately 1–1.5 hours
Less than 1% of excretion of parent drug in urine
Combination therapy with clarithromycin and amoxicillin

20 mg or 40 mg Once Daily for 4-8 Weeks for erosive esophagitis

20-80 mg Once Daily for Zollinger-Ellison Syndrome
20-40 mg Once Daily for acid-related Dyspepsia
20 mg Once Daily for 2-4 weeks for Duodenal ulcer
20-40 mg Once Daily for 4-8 weeks for Gastric ulcer
IV Injection or IV infusion dosing for specific conditions

Administration

Oral administration one hour before meals
Preparation of oral suspension
IV injection after reconstitution with 0.9% Sodium Chloride
Specific administration instructions for nasogastric or gastric tube

Interaction

Extensive metabolism in the liver by CYP2C19 and CYP3A4
