Indications

  • Peptic ulcer diseases
  • Gastroesophageal reflux diseases
  • Ulcer induced by non-steroidal anti-inflammatory drugs (NSAIDs)
  • Eradication of Helicobacter pylori (in combination with antibiotics)
  • Zollinger-Ellison Syndrome

Pharmacology

  • Proton pump inhibitor
  • Suppresses final step in gastric acid production
  • Covalently binds to H+/K+ATPase enzyme system

Dosage & Administration

  • Oral administration for benign gastric ulcer, GERD, duodenal ulcer, duodenal ulcer associated with Helicobacter pylori, prophylaxis of NSAID-associated gastric or duodenal ulcer, Zollinger-Ellison Syndrome
  • IV injection for duodenal ulcer, gastric ulcer, Gastroesophageal reflux disease associated with history of erosive esophagitis, prevention of rebleeding in peptic ulcer, prophylaxis of acid aspiration, long-term management of Zollinger-Ellison Syndrome

Interaction

  • No significant drug interactions observed in clinical studies

Contraindications

  • Known hypersensitivity to any components of the formulation

Side Effects

  • Well tolerated in short-term and long-term treatment
  • Common: headache, diarrhea
  • Rarely included: abdominal pain, flatulence, rash, insomnia, hyperglycemia

Pregnancy & Lactation

  • US FDA Pregnancy Category: B
  • No adequate and well-controlled studies in pregnant women
  • Drug excreted in human milk

Precautions & Warnings

  • Tablet should not be split, chewed or crushed
  • Long-term therapy may lead to malabsorption of cyanocobalamin (Vitamin B12)
  • May increase risk of osteoporosis related disorders
