Indications
- Symptomatic treatment of chronic constipation in adults
- Failure of previous laxative treatments
Pharmacology
- Selective stimulator of 5-HT4 receptors
- No interaction with hERG channel or 5-HT1 receptors
- Stimulation of propulsive motility in GI tract
- Release of acetylcholine
Dosage & Administration
- 2 mg once daily for adults
- 1 mg once daily for older people
- Should not be used in children under 18 years
Interaction
- Low interaction potential with other drugs
- Weak substrate for P-glycoprotein
- Potential interaction with ketoconazole and other potent inhibitors of P-gp
Contraindications
- Hypersensitivity to active substance or excipients
- Renal impairment requiring dialysis
Side Effects
- Headache
- Gastrointestinal symptoms (abdominal pain, nausea, diarrhea)
- Majority of adverse events are mild to moderate
Pregnancy & Lactation
- Not recommended during pregnancy
- Use effective contraception during treatment
- Not recommended during breastfeeding
- No indication of harmful effects in animal studies
Precautions & Warnings
- Lower dose recommended for severe renal impairment
- Caution in severe hepatic impairment
- Possible reduction in efficacy of oral contraceptives with severe diarrhea
- Contains lactose - caution for patients with rare hereditary problems of galactose intolerance
