Indications

Pharmacology
- Calcitriol as the active form of Vitamin D3
- Binding to Vitamin D receptors

Dosage and Administration
- Adult dosage: 40000 IU once weekly for 7 weeks
- Adult maintenance therapy dosage: 1400-2000 IU/day
- Children (12-18 years) dosage: 20000 IU once every 2 weeks for 6 weeks
- Film-coated tablet: 1000 IU daily
- Oroflash or chewable tablets: 1000-2000 IU daily
- Syrup dosage for patients with risk of Cholecalciferol deficiency
Interaction
- Interference of drugs with absorption and metabolism of Vitamin D3
- Drugs causing Vitamin D3 deficiencies
Contraindications
- Contradicted in diseases associated with hypercalcaemia
- Known hypersensitivity to Vitamin D3
- Evidence of Vitamin D3 toxicity

- Raised plasma and urine calcium and phosphate concentrations
Pregnancy & Lactation
- Safe use of doses up to 4000 IU during pregnancy
- Excretion of Vitamin D3 and its metabolites in breast milk
- Need to consider dose when prescribing for breast-fed infants
Precautions & Warnings
- Monitoring plasma-calcium concentration
- Consultation for patients receiving high doses
- Consultation for patients using Digoxin and Thiazide Diuretics
- Consultation for patients with liver or kidney disease, primary hyperthyroidism, lymphoma, tuberculosis, and granulomatous disease
Use in Special Populations
- Safety & efficacy in children under 12 years not established
