Pharmacology

  • Calcitriol as the active form of Vitamin D3

Dosage and Administration

  • Adult dosage: 40000 IU once weekly for 7 weeks
  • Adult maintenance therapy dosage: 1400-2000 IU/day
  • Children (12-18 years) dosage: 20000 IU once every 2 weeks for 6 weeks
  • Film-coated tablet: 1000 IU daily
  • Oroflash or chewable tablets: 1000-2000 IU daily
  • Syrup dosage for patients with risk of Cholecalciferol deficiency

Interaction

  • Interference of drugs with absorption and metabolism of Vitamin D3
  • Drugs causing Vitamin D3 deficiencies

Contraindications

  • Contradicted in diseases associated with hypercalcaemia
  • Known hypersensitivity to Vitamin D3
  • Evidence of Vitamin D3 toxicity

Pregnancy & Lactation

  • Safe use of doses up to 4000 IU during pregnancy
  • Excretion of Vitamin D3 and its metabolites in breast milk
  • Need to consider dose when prescribing for breast-fed infants

Precautions & Warnings

  • Monitoring plasma-calcium concentration
  • Consultation for patients receiving high doses
  • Consultation for patients using Digoxin and Thiazide Diuretics
  • Consultation for patients with liver or kidney disease, primary hyperthyroidism, lymphoma, tuberculosis, and granulomatous disease

Use in Special Populations

  • Safety & efficacy in children under 12 years not established

Overdose Effects

  • Hypervitaminosis D
