Indications
- Treatment of hypertension
- Reduction of stroke risk in patients with hypertension and left ventricular hypertrophy
Pharmacology
- Losartan blocks vasoconstrictor and aldosterone-secreting effects of angiotensin II
- Hydrochlorothiazide affects renal tubular mechanisms of electrolyte reabsorption
Dosage
- Usual starting dose of 50/12.5 is one tablet once daily
- Maximum dose of 100/25 once daily
- Antihypertensive effect attained within three weeks after initiation of therapy
- No initial dosage adjustment necessary for elderly patients
Administration
- May be administered with other antihypertensive agents
- May be administered with or without food
Interaction
- No significant drug-drug pharmacokinetic interactions found with certain drugs
- Possible interaction with antidiabetic drugs, other antihypertensive drugs, and cholestyramine and colestipol resins
Contraindications
- Hypersensitivity to any component of the product
- Anuria or hypersensitivity to sulfonamide-derived drugs

Precautions & Warnings
- Angioedema
- Periodic determination of serum electrolytes
- Hypokalemia may rarely develop
- Impaired renal function
- Symptomatic hypotension
Use in Special Populations
- Usual regimens of therapy may be followed with creatinine clearance >30 ml/min
- Not recommended in patients with severe hepatic impairment
- Safety and effectiveness not established in pediatric patients
Overdose Effects
- Manifestations may include hypotension, tachycardia, bradycardia
- Supportive treatment should be instituted if symptomatic hypotension occurs
