Indications
- Treatment of hypertension
- Reduction of stroke risk in patients with hypertension and left ventricular hypertrophy
Pharmacology
- Losartan as a potent vasoconstrictor
- Inhibition of aldosterone secretion by Losartan
- Renal tubular mechanisms affected by Hydrochlorothiazide
Dosage
- Usual starting dose of 50/12.5 is one tablet once daily
- Maximum dose of 100/25 once daily
- Antihypertensive effect attained within three weeks
Administration
- Can be administered with other antihypertensive agents
- Can be taken with or without food
Interaction
- No significant drug-drug pharmacokinetic interactions with certain drugs
- Possible interaction with potassium-sparing diuretics
- Potential interaction with antidiabetic drugs and other antihypertensive drugs
Contraindications
- Contraindicated in patients hypersensitive to any component
- Contraindicated in patients with anuria or hypersensitivity to sulfonamide-derived drugs
Side Effects
- Symptomatic hypotension including dizziness
- Occasional hyperkalemia
- Reports of angioedema, vertigo, gastrointestinal disturbances, and more
Pregnancy & Lactation
- Avoided in pregnancy unless essential
- Not recommended in breastfeeding
Precautions & Warnings
- Periodic determination of serum electrolytes
- Hypokalemia may rarely develop
- Monitoring required for impaired renal function and symptomatic hypotension
Use in Special Populations
- Usual regimens of therapy in patients with renal impairment
- Not recommended in patients with severe hepatic impairment
- Safety and effectiveness not established in pediatric patients
Overdose Effects
- Manifestations of overdosage could include hypotension and tachycardia
- Supportive treatment recommended for symptomatic hypotension
