Indications

  • Treatment of hypertension
  • Reduction of risk of stroke in patients with hypertension and left ventricular hypertrophy

Pharmacology

  • Losartan as a selective blocker of the binding of angiotensin II to the AT1 receptor
  • Hydrochlorothiazide as a thiazide diuretic affecting renal tubular mechanisms of electrolyte reabsorption

Dosage

  • Usual starting dose of 50/12.5 is one tablet once daily
  • Maximum dose is 100/25 once daily

Administration

  • May be administered with or without food
  • May be administered with other antihypertensive agents

Interaction

  • No significant drug-drug interactions found with specific drugs
  • Potential interactions with antidiabetic drugs, other antihypertensive drugs, and certain resins

Contraindications

  • Hypersensitivity to any component of the product
  • Anuria or hypersensitivity to sulfonamide-derived drugs

Side Effects

  • Symptomatic hypotension including dizziness
  • Occasional hyperkalaemia
  • Reported cases of angioedema, vertigo, gastrointestinal disturbances, and more

Pregnancy & Lactation

  • Avoid in pregnancy unless essential
  • Not recommended in breastfeeding

Precautions & Warnings

  • Periodic determination of serum electrolytes
  • Monitoring for hypokalemia and symptomatic hypotension

Use in Special Populations

  • Use caution in patients with renal or hepatic impairment
  • Not recommended in pediatric patients

Overdose Effects

  • Manifestations may include hypotension and tachycardia
  • Supportive treatment should be instituted for symptomatic hypotension
