Indications

Pharmacology
- Broad-spectrum parenteral cephalosporin antibiotic
- Bactericidal activity against a wide range of Gram-positive and Gram-negative organisms
- Interferes with bacterial cell wall synthesis
- High stability in the presence of beta lactamases
- Relatively long plasma elimination half-life of about 6 to 9 hours
- Not metabolized in the body
- Excreted unchanged in the urine
- Highly protein bound (95%)
Dosage
- Usual dose for adults: 1 to 2 gm intravenous or intramuscular administration once a day
- Maximum dose for adults: 4 gm/day
- Usual dose for uncomplicated gonococcal infections in adults: 250 mg intramuscular as a single dose
- Usual dose for surgical prophylaxis in adults: 1 g intravenous as a single dose
- Usual dose for infants and children: 50 to 75 mg/kg intravenous or intramuscular administration once a day
- Maximum dose for infants and children: 2 gm/day
- Duration of therapy: 4 to 14 days
Administration
- Preparation of Solutions for Intramuscular / Intravenous Injections
- Intramuscular injection solution: 250 mg or 500 mg Ceftriaxone dissolved in Lidocaine HCI 1%
- Intravenous injection solution: 250 mg or 500 mg Ceftriaxone dissolved in Water for injection

Contraindications
- Hypersensitivity to cephalosporin antibiotics
Side Effects
- Gastro-intestinal effects including diarrhea, nausea, and vomiting
- Cutaneous reactions including rash, pruritus, and urticaria
- Hematologic reactions including eosinophilia, thrombocytopenia, and anemia
- Hepatic reactions including elevations of SGOT or SGPT
- CNS reactions including nervousness, confusion, and headache
- Local phlebitis following intravenous administration
Pregnancy & Lactation
- Not established safe for human pregnancy
- Caution advised during lactation
Precautions & Warnings
- Risk of anaphylactic shock
- Potential presence of gallbladder sludge
- Requirement for regular blood picture checks during prolonged treatment
Use in Special Populations
- Must not be given to neonates if premature or newborn (up to 28 days of age)
Overdose Effects
- No specific antidote
- Treatment should be symptomatic
