Mode of Action
- Proton pump inhibitor
- Specific inhibition of the H+/K+-ATPase in gastric parietal cells
- First single optical isomer of proton pump inhibitor
- Better acid control than racemic proton pump inhibitors
- Peak plasma levels occur at approximately 1.5 hours after oral administration
- Extensively metabolized in the liver
- Plasma elimination half-life of approximately 1–1.5 hours
Dosage Form
- Oral tablets
- Oral suspension
- IV injection
- IV infusion
Dosage Instructions
- Dosage varies for different indications
- Oral dose to be taken one hour before a meal
- IV injection duration: >3 minutes, IV infusion duration: 10-30 minutes
Administration
- Tablet or capsule should be swallowed whole
- Oral suspension should be taken with water and consumed within 30 minutes
- IV injection solution to be administered intravenously over a period of at least 3 minutes
Interactions
- Extensively metabolized by CYP2C19 and CYP3A4
- No clinically significant interactions with certain drugs
- Has potential to interfere with the absorption of drugs where gastric pH is important for bioavailability
Contraindications
- Known hypersensitivity to any of the formulation

Pregnancy & Lactation
- No adequate and well-controlled studies in pregnant women

- Symptomatic response to therapy does not preclude the presence of gastric malignancy
- Dosage adjustments for hepatic insufficiency
- No dosage adjustment recommended for renal insufficiency
Use in Special Populations
- Safety and effectiveness in pediatric patients not established
- No overall differences in safety and efficacy observed between the elderly and younger individuals
Overdose Effects
- Toxic dose lethal to rats
- Major signs include reduced motor activity, changes in respiratory frequency, tremor, ataxia, and intermittent clonic convulsions

- Store at a temperature not exceeding 30°C
- Protect from light and moisture
- Keep out of reach of children
