Indications
- Neuropathic pain associated with diabetic peripheral neuropathy (DPN)
- Postherpetic neuralgia (PHN)
- Adjunctive therapy for the treatment of partial-onset seizures in patients 1 month of age and older
- Fibromyalgia
- Neuropathic pain associated with spinal cord injury
Pharmacology
Structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA)
Dosage
- DPN in adults: 50-300 mg/day
- PHN in adults: 75-600 mg/day
- Fibromyalgia in adults: 300-450 mg/day
- Neuropathic pain associated with spinal cord injury in adults: 150-600 mg/day
Administration
- Route of administration: Oral
- Dosage form: CR tablet
- Administration instructions: Swallow whole, do not split, crush, or chew
- Discontinuation: Gradual tapering over a minimum of 1 week

- Drug interaction with medication: Unlikely significant pharmacokinetic interactions
- Drug interaction with food and others: Not applicable
Contraindications
Known hypersensitivity to Pregabalin or its components

- Most common in adults:
- Dizziness
- Somnolence
- Dry mouth
- Edema
- Blurred vision
- Weight gain
- Thinking abnormal
- Most common in pediatric patients:
- Increased weight
- Increased appetite
Pregnancy & Lactation
No adequate and well-controlled studies, detected in the milk of lactating women, not recommended during breastfeeding
Precautions & Warnings
- Angioedema can occur and may be associated with life threatening respiratory compromise
- Discontinue immediately in case of hypersensitivity reactions
- Increase in suicidal thoughts or behavior
- Respiratory depression may occur
- May cause dizziness and somnolence, impairing ability to drive or operate machinery
- Caution when coadministering with thiazolidinedione antidiabetic agents
Use in Special Populations
Safety and effectiveness in pediatric patients have not been established
Overdose Effects
Reduced consciousness, depression/anxiety, seizures, heart block, no specific antidote
