Losart Plus 50 mg+12.5 mg (Tablet)

Indications

• Treatment of hypertension
• Reduction of stroke risk in patients with hypertension and left ventricular hypertrophy

Pharmacology

• Actions of losartan and hydrochlorothiazide on renin-angiotensin system
• Affect on vasoconstriction and aldosterone secretion
• Inhibition of AT1 receptor
• Diuretic action of hydrochlorothiazide

Dosage

• Usual starting dose of 50/12.5 once daily
• Max dose of 100/25 once daily
• Antihypertensive effect within three weeks
• No initial dosage adjustment for elderly patients

Administration

• May be administered with other antihypertensive agents
• May be administered with or without food

Interaction

• No significant drug-drug interactions with losartan and hydrochlorothiazide
• Potential interactions with potassium-sparing diuretics, non-steroidal anti-inflammatory drugs, alcohol, barbiturates, narcotics, antidiabetic drugs, other antihypertensive drugs, cholestyramine, and colestipol resins

Contraindications

• Hypersensitivity to any component
• Anuria or hypersensitivity to sulfonamide-derived drugs

Pregnancy & Lactation

• Avoid in pregnancy unless essential
• Adverse effects on fetal and neonatal blood pressure control and renal function
• Not recommended in breastfeeding

Precautions & Warnings

• Hypersensitivity: Angioedema
• Periodic determination of serum electrolytes
• Hypokalemia
• Impaired renal function
• Symptomatic hypotension

Use in Special Populations

• Usual regimens for patients with renal impairment
• Not recommended in patients with severe renal impairment
• Not recommended in patients with hepatic impairment
• Not established safety and effectiveness in pediatric patients

Overdose Effects

• Manifestations of losartan and hydrochlorothiazide overdosage
• Supportive treatment for symptomatic hypotension
• Removal by hemodialysis not applicable for losartan or its metabolite
• Signs and symptoms caused by electrolyte depletion
