Indications

  • Treatment of hypertension
  • Reduction of stroke risk in patients with hypertension and left ventricular hypertrophy

Pharmacology

    • Angiotensin II Effects: Vasoconstriction, aldosterone secretion
    • Losartan Action: Blocks vasoconstrictor and aldosterone-secreting effects of angiotensin II by selectively blocking AT1 receptor
    • Hydrochlorothiazide Action: Affects renal tubular mechanisms of electrolyte reabsorption, increases excretion of Sodium and Chloride

Dosage

  • Usual Starting Dose: 50/12.5 - one tablet once daily
  • Maximum Dose: 100/25 once daily
  • Severe Hypertension Initial Treatment: 50/12.5 once daily, may be increased to 100/25 once daily

Administration

May be administered with or without food, may be used with other antihypertensive agents

Interaction

  • Losartan Potassium: No significant drug-drug interactions found, potassium-sparing diuretics may lead to increased serum potassium
  • Hydrochlorothiazide: May interact with alcohol, barbiturates, narcotics, antidiabetic drugs, and other antihypertensive drugs

  • Hypersensitivity to any component of the product
  • Anuria
  • Hypersensitivity to sulfonamide-derived drugs

  • Symptomatic hypotension

Use in Special Populations

  • Use in Patients with Renal Impairment
  • Use in Patients with Hepatic Impairment
  • Use in pediatric patients not established

Overdose Effects

  • Losartan Potassium: Manifestations may include hypotension and tachycardia
  • Hydrochlorothiazide: Signs and symptoms caused by electrolyte depletion
