Description

  • Benzimidazole anthelmintic for single and mixed infestations of various parasites.

Indications

  • Indicated in infestations of hookworm, roundworm, threadworm, whipworm, strongyloides, tapeworm, opisthorchi, and hydatid.

Pharmacology

  • Broad spectrum anthelmintic with vermicidal, ovicidal, and larvicidal activities. Thought to exert its anthelmintic effect by blocking glucose uptake in helminths.

Mode of action

  • Inhibitory effect on tubulin polymerization resulting in the loss of cytoplasmic microtubules. Metabolized in the liver with a plasma half-life of about 8.5 hrs. Excreted in the urine.

Dosage administration

  • Recommended dosages for adults, children over 2 years, children of 1-2 years, and those under 1 year. Specific dosages for hydatid disease, strongyloidiasis, taeniasis, cystic echinococcosis, alveolar echinococcosis, and giardiasis.

Interaction

  • No reported pharmacodynamic or pharmacokinetic interaction.

Contraindications

  • Not recommended for use in neonates, pregnant women, and children weighing less than 10 kg. No evidence to suggest dose alteration in renal, hepatic, or cardiac failure.

Side effects

  • Potential gastrointestinal disturbances, headache, dizziness, changes in liver enzymes, reversible alopecia, rash, fever, blood disorders, allergic shock, convulsion, and meningism.

Pregnancy lactation

  • FDA Pregnancy category C. Should be avoided in pregnancy and lactation unless the potential benefits outweigh the possible risks to the fetus.

Precautions warnings

  • Blood counts and liver function tests before treatment and during each cycle. Considerations for breastfeeding, exclude pregnancy before starting treatment. Use in Echinococcosis necessitates constant medical supervision and monitoring.
