Indications
- Palliative treatment of advanced carcinoma of the breast or endometrium
- Treatment of anorexia, cachexia, or unexplained weight loss in patients with AIDS & cancer
Pharmacology
- Synthetic, antineoplastic and progestational drug
- Inhibition of pituitary gonadotrophin production
- Modifying and abolishing the stimulatory effects of estrogen on cells
- Plasma half-life of 33 to 38 hours
- Peak plasma concentrations 2 to 3 hours after a single oral dose
Dosage & Administration
- Tablet dosage for breast cancer: 160 mg/day
- Tablet dosage for endometrial carcinoma: 40-320 mg/day in divided doses
- Oral suspension recommended initial dosage: 800 mg/day
Interaction
- Possible interactions with concomitant medications not investigated
- Estrogens and Progestogens may cause fluid retention
Contraindications
- History of hypersensitivity to Megestrol Acetate or any component
- Known or suspected pregnancy
Side Effects
- Weight gain associated with increased appetite
- Thromboembolic phenomena including thrombophlebitis and pulmonary embolism
- Clinical cases of new onset diabetes mellitus
- Nausea, dyspnea, tumor flare, hyperglycemia, glucose intolerance, alopecia, hypertension
- Carpal tunnel syndrome, mood changes, hot flashes, malaise, asthenia, lethargy, sweating and rash
Pregnancy & Lactation
- Pregnancy Category D
- Small amounts excreted in mother's milk
- Nursing should be discontinued during treatment with Megestrol Acetate
Precautions & Warnings
- Close surveillance for patients treated for recurrent or metastatic cancer
- Use with caution in patients with history of thromboembolic disease
- Exacerbation of preexisting diabetes with increased insulin requirements
- Insufficient data for patients 65 years and older
Use in Special Populations
- Safety and effectiveness in pediatric patients not established
- Insufficient data for patients 65 years and older
Overdose Effects
- No serious unexpected side effects at dosages as high as 1600 mg/day
