Indications
- Invasive aspergillosis
- Candidemia (nonneutropenic)
- Disseminated candidiasis
- Esophageal candidiasis
- Serious infections caused by Scedosporium apiospermum
- Intolerant of other therapy
Pharmacology
- Triazole antifungal medication
- Binds and inhibits ergosterol synthesis
- Inhibits CYP450-dependent 14-alpha sterol demethylase
- Results in depletion of ergosterol in fungal cell membrane
Dosage & Administration
- Oral administration
- To be taken at least one hour before or after a meal
- Different dosages for patients above and below 40 kg body weight
- Loading dose regimen and maintenance dose for different conditions
- Injection administration for invasive aspergillosis, candidemia, scedosporiosis, and fusariosis
Interaction
- Requires dosage adjustment with CYP3A4, CYP2C9, and CYP2C19 inhibitors/inducers
- May increase concentrations and activity of other drugs
- Specific dose adjustments with co-administration of Phenytoin or Efavirenz
Contraindications
- Known hypersensitivity to Voriconazole
- Co-administration with specific drugs due to risk of adverse reactions or loss of efficacy
