Indications
- Treatment of invasive aspergillosis
- Treatment of candidemia (nonneutropenic) and disseminated candidiasis
- Treatment of esophageal candidiasis
- Treatment of serious infections caused by Scedosporium apiospermum and Fusarium Species
- Use in patients intolerant of, or refractory to other therapy
Pharmacology
- Voriconazole is a triazole antifungal medication
- Binds and inhibits ergosterol synthesis
- Inhibits CYP450-dependent 14-alpha sterol demethylase
- Results in depletion of ergosterol in fungal cell membrane
Dosage & Administration
- Oral administration - tablets and powder for suspension
- Loading dose regimen for body weight over and below 40 kg
- Maintenance dose regimen
- Injection dosage for invasive aspergillosis, candidemia, Scedosporiosis and Fusariosis
Interaction
- Adjustment of Voriconazole dosage with CYP3A4, CYP2C9 and CYP2C19 inhibitors and inducers
- Increase in concentrations and activity of other drugs with Voriconazole
- Alteration of oral and intravenous dosage with co-administration of Phenytoin or Efavirenz
Contraindications
- Hypersensitivity to Voriconazole or any other components
- Co-administration with specific drugs due to risk of adverse reactions or loss of efficacy
