Indications

  • Treatment of partial-onset seizures with or without secondary generalization in adults and adolescents
  • Treatment of myoclonic seizures in adults and adolescents
  • Treatment of primary generalized tonic-clonic seizures in adults and adolescents

Description

  • Precise mechanism not fully understood
  • Minimal activity in submaximal stimulation and threshold tests
  • Inhibition of burst firing without affecting normal neuronal excitability

Dosage

  • Recommended starting dose for adults and adolescents from 16 years of age
  • Maximum dose for adults and adolescents (≥18 years) weighing 50 kg or more
  • Tablet formulation not adapted for use in infants and children under the age of 6 years

Administration

  • Oral administration with sufficient quantity of liquid
  • Daily dose administered in two equally divided doses

Interaction

  • Data on interaction with alcohol not available
  • Inhibition of renal clearance of primary metabolite by probenecid
  • Concomitant administration with methotrexate can decrease methotrexate clearance

Contraindications

  • Hypersensitivity to active substance or other pyrrolidone derivatives

  • Most common adverse reactions include somnolence and asthenia in adults

Pregnancy & Lactation

  • Not recommended during pregnancy and lactation

  • Rare cases of decreased blood cell counts reported
  • Suicidal ideation and behaviour reported

Use in Special Populations

  • Dose adjustment recommended for elderly patients with compromised renal function
  • Daily dose must be individualized according to renal function

Overdose Effects

  • Symptomatic treatment may include haemodialysis
