Indications
- Monotherapy for partial-onset seizures in adults and adolescents
- Adjunctive therapy for various types of seizures in adults, adolescents, children, and infants
Description
- Mechanism of antiepileptic effect is unknown
- Minimal activity in submaximal stimulation and threshold tests
- Protection against secondarily generalized activity from induced seizures
- Inhibition of burst firing without affecting normal neuronal excitability
Dosage
- Recommended starting dose for adults and adolescents from 16 years of age
- Initial therapeutic dose for adults (≥18 years) and adolescents (12 to 17 years) weighing 50 kg or more
- Use of oral solution for pediatric population
- No established safety and efficacy for monotherapy in children and adolescents below 16 years
Administration
- Oral administration in two equally divided doses
Interaction
- No influence on serum concentrations of existing antiepileptic medicinal products
- Inhibition of renal clearance of methotrexate when administered concomitantly
- Reports of decreased efficacy when administered with osmotic laxative macrogol
Contraindications
- Hypersensitivity to the active substance or other pyrrolidone derivatives or to any of the excipients
Side Effects
- Common adverse reactions in adult and pediatric patients
- Incidence of somnolence, asthenia, infection, dizziness in adult patients
- Incidence of fatigue, aggression, nasal congestion, decreased appetite, irritability in pediatric patients
Pregnancy & Lactation
- Not recommended during pregnancy and lactation
- Excretion in human breast milk
Precautions & Warnings
- Dose adjustment for renal impairment
- Rare cases of decreased blood cell counts
- Risk of suicidal thoughts and behavior
- Tablet formulation not adapted for use in infants and children under 6 years
Use in Special Populations
- Recommended dose adjustment in elderly patients with compromised renal function
- Individualized daily dose according to renal function in patients with renal impairment
- No dose adjustment needed in patients with mild to moderate hepatic impairment
Overdose Effects
- Adverse effects observed with overdose
- Symptomatic treatment may include haemodialysis
- No specific antidote available
