Indications

  • Invasive aspergillosis
  • Candidemia (nonneutropenic)

  • Intolerance or refractory to other therapy

Pharmacology

  • Triazole antifungal medication
  • Binds and inhibits ergosterol synthesis
  • Inhibits CYP450-dependent 14-alpha sterol demethylase

DosageAdministration

  • Loading and maintenance doses based on body weight

Interaction

  • CYP3A4, CYP2C9 and CYP2C19 inhibitors and inducers affect dosage
  • Increase in concentrations and activity of CYP3A4, CYP2C9 and CYP2C19 substrates
  • Co-administration with Phenytoin or Efavirenz affects dosage
