Indications

  • Primary hyperlipidemia and mixed dyslipidemia
  • Hypertriglyceredemia
  • Primary dysbetalipoproteinemia
  • Homozygous familial hypercholesterolemia (HoFH)
  • Atherosclerosis progression reduction
  • Risk reduction of Myocardial Infarction (MI), stroke, and arterial revascularization procedures
  • Pediatric patients 10 to 17 years of age with heterozygous familial hypercholesterolemia (HeFH)

Pharmacology

  • Selective and competitive inhibitor of HMG-CoA reductase
  • Increases hepatic LDL receptors
  • Inhibits hepatic synthesis of VLDL and LDL particles

Dosage & Administration

  • 5-20 mg/day for pediatric patients (10 to 17 years of age)
  • 40 mg once daily for very high cholesterol and high risk of MI or stroke
  • 20 mg once daily for homozygous familial hypercholesterolemia
  • Initiation with 5 mg once daily for Asian patients
  • Limited dose with concomitant use of Cyclosporine, Lopinavir/Ritonavir, Atazanavir/Ritonavir, Niacin, Fenofibrate, and Gemfibrozil
  • Dosing adjustment for patients with severe renal impairment

  • Known hypersensitivity to product components

Pregnancy & Lactation

  • Monitoring of liver enzyme tests
  • Discontinue therapy if markedly elevated creatinine kinase levels occur
