Indications
- Treatment of nausea and vomiting of pregnancy
- For women who do not respond to conservative management
Pharmacology
- Doxylamine Succinate is an antihistamine
- Blocks Histamine (H1) receptor
- Crosses blood-brain barrier
- Decreases action of histamine at H1 receptor
- Affects vestibular system
- Decreases stimulation of vomiting center
- Pyridoxine Hydrochloride is a vitamin B6 analog
- Used to prevent nausea and vomiting
- Antiemetic properties
Dosage & Administration
- Initial dosage of one tablet orally at bedtime
- Maximum recommended dose of two tablets per day
- Take on an empty stomach with water
- Swallow tablets whole
- Do not crush, chew, or split tablets
- Take daily and not on as needed basis
Interaction
- Contraindicated with monoamine oxidase inhibitors (MAOIs)
- Not recommended with alcohol and other CNS depressants
Contraindications
- Hypersensitivity to Doxylamine Succinate or Pyridoxine Hydrochloride
- Concurrent use of MAO inhibitors

- Somnolence
- Accidents resulting from combined use with CNS depressants

- Pregnancy category A
- Excreted into breast milk
- Caution while breastfeeding
Precautions & Warnings
- May cause somnolence
- Avoid activities requiring alertness
- Not recommended with concurrent use of CNS depressants
- Use with caution in women with asthma, increased intraocular pressure, narrow angle glaucoma, stenosing peptic ulcer, pyloroduodenal obstruction, or urinary bladder-neck obstruction
Overdose Effects
- Extended-release or Delayed Release formulation
- Signs and symptoms may not be apparent immediately
- Signs and symptoms may include restlessness, dry mouth, dilated pupils, sleepiness, vertigo, mental confusion, tachycardia
- At toxic doses, exhibits anticholinergic effects, seizures, rhabdomyolysis, acute renal failure, death
- Treatment includes gastric lavage, activated charcoal, whole bowel irrigation, symptomatic treatment
