Mantadin 100 mg (Capsule)

Indications

• Treatment of parkinsonism
• Treatment of drug-induced extrapyramidal reactions
• Prophylaxis and treatment of signs and symptoms of infection caused by various strains of influenza A virus

Pharmacology

• Prevents the release of infectious viral nucleic acid into the host cell
• Interferes with the function of the transmembrane domain of the viral M2 protein
• Inhibits the replication of influenza A virus isolates from H1N1, H2N2 and H3N2 subtypes

Dosage & Administration

• Adult dose for parkinson's disease: Initially 100 mg/day, increased to 100 mg bid after a week or more
• Adult dose for prophylaxis of influenza A: 100 mg daily for up to 6 weeks

Interaction

• Concurrent administration with anticholinergic agents or levodopa may increase confusion, hallucinations, or other atropine-like side effects
• Additive CNS toxicity may result from concurrent administration with drugs or substances acting on the CNS

Contraindications

• Hypersensitivity to the active substances or to any of the excipients

• Nausea
• Dizziness/lightheadedness
• Insomnia
• Edema of ankles
• Livedo reticularis
• Anxiety
• Hallucinations
• Blurred vision

Pregnancy & Lactation

• Pregnancy category C
• Excreted into breast milk in low concentrations
• Use cautiously in women who are breastfeeding

Precautions & Warnings

• Should not be discontinued abruptly in patients with Parkinson's disease
• Dose reduction of anticholinergic drugs or of Mantadin is necessary if atropine-like effects appear

Use in Special Populations

• Patients with renal impairment: Dose reduction is required based on creatinine clearance
• Patients with hepatic impairment: Use with caution
