Description
- Tricyclic compound
- Antagonizes serotonin receptor competitively
- Antihistamine properties

Pharmacology
- Inhibits peripheral actions of serotonin and histamine
- Increases membrane permeability of cranial vessels
- Alters pain thresholds in migraines
- Blocks 5-HT receptors
- Attenuates signaling of serotonin
- Inhibits peripheral actions of bradykinin
- Inhibits serotonin reuptake by blood platelets
- Affects tonicity and decreases passive distension of extracranial arteries
- Stimulates appetite at metabolic level
Dosage & Administration
- Recommended dosage for adults: 1.5 mg daily
- Can be taken as a single dose at night or in three divided doses
- Maximum dosage for adults: 4.5 mg daily
- Up to 3 mg may be given as a single dose
- Recommended dosage for children (aged over 2 years): Up to 1.5 mg daily
- Maximum dosage for children: Up to 1 mg as a single dose at night
Interaction
- Enhanced central effects of sedatives, hypnotics, antihistamines, and alcohol
- Antagonizes hypotensive effect of adrenergic neurone blockers
Contraindications
- Contraindicated in patients with known hypersensitivity to any of its ingredients
