Indications

Treatment of nausea and vomiting of pregnancy

Pharmacology

Doxylamine Succinate as antihistamine
Blocks Histamine (H1) receptor
Crosses blood brain barrier
Decreases action of histamine at H1 receptor
Inhibition of vestibular system & Muscarinic receptor
Effects on vestibular system
Pyridoxine Hydrochloride as a vitamin B6 analog
Prevents nausea and vomiting

Dosage & Administration

Initial dosage is one tablet orally at bedtime
Maximum recommended dose is two tablets per day
Take on an empty stomach with a glass of water
Swallow tablets whole
Daily usage

Interaction

Contraindicated with monoamine oxidase inhibitors (MAOIs)
Not recommended with alcohol and other CNS depressants

Hypersensitivity to Doxylamine Succinate or Pyridoxine Hydrochloride
Use with MAO inhibitors

Overdose Effects

Extended-release or Delayed Release formulation
Signs and symptoms of intoxication may not be immediate
Symptoms of overdose include restlessness, dry mouth, dilated pupils, sleepiness, vertigo, mental confusion, tachycardia
May exhibit anticholinergic effects at toxic doses
Treatment involves gastric lavage, activated charcoal, whole bowel irrigation, and symptomatic treatment
