Indications

Pharmacology
- Active form: Calcitriol
- Binds to Vitamin D receptors (VDRs)
- Half-life of about 50 days
- Fat soluble
- Absorbed in the small intestine
- Metabolized to 25-hydroxy Vitamin D3 (Calcidiol)
- Second hydroxylation to 1,25-dihydroxy Vitamin D3 (Calcitriol) in the kidney
- Increases calcium absorption
- Stored in tissues such as fat and muscle
- Eliminated via faeces and urine
Dosage & Administration
- Adult dosage: 40000 IU once weekly for 7 weeks
- Maintenance therapy dosage: 1400-2000 IU/day
- Children (12-18 years) dosage: 20000 IU once every 2 weeks for 6 weeks
- Daily tablet dosage: 1000 IU (1-2 tablets)
- Syrup dosage for patients with risk of Cholecalciferol deficiency
Interaction
- Drugs causing Vitamin D3 deficiencies
- Interference with absorption and metabolism
- Examples: Magnesium containing Antacids, Digoxin, Thiazide Diuretics, Cholestyramine, Corticosteroids
Contraindications
- Hypercalcaemia
- Hypersensitivity to Vitamin D3
- Evidence of Vitamin D3 toxicity

- Safe use up to 4000 IU during pregnancy
- Recommended daily intake for pregnant women: 400 IU
- Excretion in breast milk
- No observed overdose in nursing infants
