Indications
- Gastric and duodenal ulcer
- NSAID-associated duodenal and gastric ulcer

Pharmacology
- Inhibitor of gastric acid secretion
- Blocks hydrogen-potassium-adenosine triphosphatase (H+/K+ ATPase) enzyme system
- Onset of the antisecretory effect occurs within one hour
- Maximum effect within two hours
- Inhibition of secretion lasts up to 72 hours
- Secretory activity returns gradually over 3 to 5 days after discontinuation

Interaction

Contraindications
- Known hypersensitivity to omeprazole
- Contraindicated if gastric ulcer is suspected and malignancy needs to be excluded
Side Effects
- Mild and reversible adverse reactions
- Reported skin reactions like rash, urticaria, and pruritus
- Digestive system reactions such as diarrhoea, headache, constipation, and nausea
- Neurological reactions like dizziness, headache, somnolence, and insomnia
- Respiratory reactions like bronchospasm
- Hematological reactions like leucopenia, thrombocytopenia, and pancytopenia
- Hepatic reactions like increases in liver enzymes
- Other reactions like photosensitivity, alopecia, dry mouth, and peripheral oedema
Pregnancy & Lactation
- No adverse effects of omeprazole on pregnancy or fetus/newborn child
- Use during pregnancy is possible
- No information on passage into breast milk or effects on the neonate
Precautions & Warnings
- Avoid concomitant use of clopidogrel and Lovanzo
- Possible risk of osteoporosis-related fractures with PPI therapy
- Atrophic gastritis noted in patients treated long-term with Lovanzo
- Potential methotrexate toxicities with PPIs
