Pharmacology

  • Inhibitor of gastric acid secretion

  • Inhibition of secretion lasts up to 72 hours
  • Secretory activity returns gradually over 3 to 5 days after discontinuation

Contraindications

  • Known hypersensitivity to omeprazole
  • Contraindicated if gastric ulcer is suspected and malignancy needs to be excluded

Side Effects

  • Mild and reversible adverse reactions
  • Reported skin reactions like rash, urticaria, and pruritus
  • Digestive system reactions such as diarrhoea, headache, constipation, and nausea
  • Neurological reactions like dizziness, headache, somnolence, and insomnia
  • Respiratory reactions like bronchospasm
  • Hematological reactions like leucopenia, thrombocytopenia, and pancytopenia
  • Hepatic reactions like increases in liver enzymes
  • Other reactions like photosensitivity, alopecia, dry mouth, and peripheral oedema

Pregnancy & Lactation

  • No adverse effects of omeprazole on pregnancy or fetus/newborn child
  • Use during pregnancy is possible
  • No information on passage into breast milk or effects on the neonate

Precautions & Warnings

  • Avoid concomitant use of clopidogrel and Lovanzo
  • Possible risk of osteoporosis-related fractures with PPI therapy
  • Atrophic gastritis noted in patients treated long-term with Lovanzo
  • Potential methotrexate toxicities with PPIs
