Indications and Usage
- Symptomatic treatment of chronic constipation in adults
- Laxative failure relief
Pharmacology
- Selective stimulator of 5-HT4 receptors
- No interaction with hERG channel or 5-HT1 receptors
- Stimulates motility by releasing acetylcholine in the GI tract
Dosage and Administration
- Adults: 2 mg once daily with or without food
- Older people: Start with 1 mg once daily
- Children: Not to be used in children and adolescents younger than 18 years
Interactions
- Low interaction potential
- Not expected to affect CYP-mediated metabolism of co-medicated medicinal products
- Weak substrate for P-glycoprotein
Contraindications
- Hypersensitivity to active substance or excipients
- Renal impairment requiring dialysis

Pregnancy and Lactation
- Not recommended during pregnancy
- Use effective contraception during treatment
- Not recommended during breastfeeding
Precautions and Warnings
- Dose adjustment in severe renal impairment

- Possible reduction in efficacy of oral contraceptives in case of severe diarrhea

- Renal impairment: Dose adjustment based on GFR
- Hepatic impairment: Dose adjustment for severe hepatic impairment
- No dose adjustment for mild to moderate impairment
Overdose Effects
- Symptomatic treatment and supportive measures
- Possible symptoms: headache, nausea, diarrhea
- Correction of electrolyte disturbances if extensive fluid loss
