Valestra 1 mg (Tablet)

Indications

• Hormone replacement therapy (HRT) for estrogen deficiency symptoms in peri- and postmenopausal women
• Prevention of osteoporosis in postmenopausal women at high risk of future fracture

Pharmacology

• Estradiol enters target cells freely and interacts with a target cell receptor
• Estrogens increase hepatic synthesis of sex hormone binding globulin (SHBG) and other serum proteins
• Metabolized by cytochrome P450

Dosage & Administration

• 1 mg tablet to be taken daily
• Continuous treatment without a break
• Lowest effective dose for the shortest duration should be used

Contraindications

• Known, past, or suspected breast cancer
• Undiagnosed genital bleeding
• Previous idiopathic or current venous thromboembolism

Side Effects

• Breast cancer
• Abnormal growth or cancer of the lining of the womb
• Ovarian cancer
• Blood clots in the veins of the legs or lungs
• Heart disease
• Stroke
• Memory loss if HRT is started over the age of 65

Pregnancy & Lactation

• Not indicated during pregnancy
• Treatment should be withdrawn immediately if pregnancy occurs
• No teratogenic or foetotoxic effects indicated by most epidemiological studies

Precautions & Warnings

• HRT should only be initiated for symptoms that adversely affect quality of life
• Careful appraisal of risk and benefits should be undertaken annually
• Complete personal and family medical history should be taken before initiating or reinstituting HRT

Overdose Effects

• Nausea and vomiting
• Withdrawal bleeding may occur in some women
• No specific antidotes available
