Indications
- Used for the treatment of hypertension
- Lowers blood pressure
- Reduces the risk of cardiovascular events
Pharmacology
- Dihydropyridine calcium antagonist
- Inhibits transmembrane influx of calcium ions
- Greater effect on vascular smooth muscle cells than on cardiac muscle cells
- Pharmacologically active isomer: levamlodipine
- Absorption details: Tmax, bioavailability, unaffected by food
- Distribution: volume of distribution similar to amlodipine
- Metabolism: 90% metabolized to inactive metabolites, primarily mediated by CYP3A4
- Excretion: 60% eliminated in urine, 10% as unmetabolized drug
Dosage & Administration
- Oral route of administration
- Usual initial antihypertensive oral dose: 2.5 mg once daily
- Maximum dose: 5 mg once daily
- Dosage for adult and elderly patients
- Dosage for children and adolescents
Interaction

Contraindications
- Contraindicated in patients with known sensitivity to amlodipine

- Rare side effects: diarrhea, back pain, headache, fatigue, nausea
Pregnancy & Lactation
- Limited data on amlodipine use in pregnant women
- No adverse effects on the breastfed infant observed
Precautions & Warnings
- Symptomatic hypotension
- Worsening angina or acute myocardial infarction
- Administration in patients with severe hepatic impairment
Overdose Effects
- Excessive peripheral vasodilation with marked hypotension, reflex tachycardia possible
