Indications
- Prevention and treatment of angina pectoris due to coronary artery disease
Pharmacology
- Relaxation of vascular smooth muscle
- Dilatation of peripheral arteries and veins
- Reduction of left ventricular end-diastolic pressure
- Dilation of the coronary arteries
- Reduction of systemic vascular resistance
Dosage & Administration
- Oral dose of 1 tablet (20 mg) 2 times daily
- Maintenance doses ranging from 20 mg to 120 mg
- Oral dose of 1 capsule (50 mg) daily
Interaction
- Contraindicated with phosphodiesterase inhibitors
- Contraindicated with riociguat
- Additive effects with other vasodilators
- Possible need for dose adjustments when used with calcium channel blockers

Overdose Effects
- Symptoms include hypotension, throbbing headache, tachycardia, flushing
- Methemoglobinemia may occur
- Treatment with recumbent position, fluids, alpha-adrenergic vasopressors, methyline blue
