Usability Features
- Should be taken 15-30 minutes before meals
Dosage & Administration
- Adults: 10-20 mg (1-2 tablets or 10-20 ml suspension) every 6-8 hours daily
- Children: 2-4 ml suspension/10 kg body weight every 6-8 hours daily
Interaction
- Concomitant administration of anticholinergic drugs may antagonize the antidyspeptic effect of domperidone
- Antacids and antisecretory drugs should not be given simultaneously with domperidone
- Main metabolic pathway through CYP3A4
- In vitro data suggest concomitant use of drugs that significantly inhibit CYP3A4 may increase plasma levels of domperidone
Contraindications
- Known hypersensitivity to domperidone
- Neonates
- Gastrointestinal hemorrhage, mechanical obstruction or perforation
- Prolactin releasing pituitary tumor (prolactinoma)
Side Effects
- Transient intestinal cramps
- Rare extrapyramidal phenomena
- Increase in plasma prolactin level
- Rare allergic reactions such as rash and urticaria
Pregnancy & Lactation
- Should only be used during the first trimester of pregnancy if justified by therapeutic benefit
- Not known whether excreted in breast milk is harmful to the newborn
- Nursing not recommended unless expected benefits outweigh potential risk
Precautions & Warnings
- Absolute caution in children due to increased risk of extrapyramidal reactions
- Use with caution in patients with hepatic impairment
Special Populations
- Use with caution in infants due to immature blood-brain barrier
- Use with caution in patients with hepatic impairment
- Dosing frequency may need to be reduced in patients with severe renal insufficiency
Overdose Effects
- Symptoms may include drowsiness, disorientation, and extrapyramidal reactions
- Administration of activated charcoal and close observation recommended
