Composition
- Thiopental 500 mg injection
- Thiopental 1 gm injection
Pharmacology
- Ultrashort-acting depressant of the central nervous system
- Binds at GABAA receptor
- Produces hypnosis within 30 to 40 seconds
- Rapid recovery after small dose
- Prolonged anesthesia with repeated doses
Dosage & Administration
- Administered intravenously as 2.5% w/v solution
- Premedication with atropine or scopolamine
- Test dose for tolerance assessment
- Normal dosage for induction of anesthesia
- Dosage considerations for adults, children, and elderly
- Use in convulsive states
Interaction
- Possible increase in difficulty in producing anesthesia with alcohol or CNS depressants
- Additive action with other CNS depressants
- Hypotension and excitatory effects with phenothiazine antipsychotics
- Decreased requirement of Thiontal with certain drugs
Contraindications
- Absence of suitable veins for intravenous administration
- Hypersensitivity to barbiturates
- Porphyria
- Status asthmaticus
- Severe cardiovascular disease
- Hypotension or shock
- Conditions prolonging or potentiating hypnotic effect

Overdose Effects
- Too rapid or repeated administration may lead to overdose
- Alarming fall in blood pressure
- Respiratory difficulties and laryngospasm
- Discontinue in case of suspected overdosage
