Description
- Indications:
- Short-term treatment of serious infections due to susceptible strains of Gram-negative bacteria
- Effective in bacterial septicemia, serious respiratory tract infections, bone and joint infections, central nervous system infections, skin and soft tissue infections, intra-abdominal infections, burns, postoperative infections, complicated and recurrent urinary tract infections
- Effective in infections caused by gentamicin and/or tobramycin-resistant strains of Gram-negative organisms
- Effective in staphylococci infections
- Pharmacology:
- Semi-synthetic aminoglycoside antibiotic
- Active against pseudomonas species, Escherichia coli, Proteus species, Providencia species, Klebsiella-Enterobacter species, Acinetobacter species, and Citrobacter freundii
- Active against penicillinase and nonpenicillinase-producing Staphylococcus species including methicillin-resistant strains
- Dosage:
- Adult dosage of 15 mg/kg/day in two equally-divided doses
- Neonates and premature children dosage with an initial loading dose of 10 mg/kg followed by 15 mg/kg/day in two equally divided doses
- Elderly dosage adjusting as per impaired renal function
- Increased adult dose for life-threatening infections and/or those caused by Pseudomonas
- Dosage for urinary tract infections and impaired renal function
- Administration:
- Preferred intramuscular route for most infections
- Intravenous route for life-threatening infections or when intramuscular route is not feasible
- Intraperitoneal use as an irrigant after recovery from anesthesia in concentration of 0.25%
- Interaction:
- Concurrent administration with myorelaxants potentiate their effects
- Avoid combination with other Aminoglycoside antibiotics and fast acting diuretics
- Increased risk of ototoxicity with concurrent administration with cephalosporins or polymixins
- Contraindications:
- Contraindicated in patients with known hypersensitivity to Amikacin or any constituents of the injection
- Side Effects:
- Adverse effects include tinnitus, vertigo, reversible or irreversible deafness, skin rash, drug fever, headache, paraesthesia, nausea and vomiting
- Pregnancy & Lactation:
- Rapid crossing of the placenta into the fetal circulation and amniotic fluid
- Potential risk of ototoxicity in the fetus
- No information available regarding safety during breastfeeding
- Precautions & Warnings:
- High concentrations in the renal excretory system, requires patients to be well hydrated to minimize chemical irritation of the renal tubules
- Monitoring of renal function during treatment is important
- Use in Special Populations:
- Safety and effectiveness in children or adolescents under 16 years not established
- Overdose Effects:
- Peritoneal or haemodialysis advised in the event of overdose or toxic reaction
