Indications

  • Essential hypertension
  • Angina pectoris & hypertension co-existing diseases
  • Post Myocardial Infarction (MI) patients
  • Refractory angina pectoris with failed nitrate therapy

Pharmacology

  • Fixed-dose combination of Amlodipine and Atenolol
  • Amlodipine is a dihydropyridine calcium antagonist
  • Amlodipine acts as a peripheral vasodilator
  • Atenolol is a cardioselective beta-blocker
  • Atenolol causes reduction in blood pressure and myocardial oxygen demand

Dosage & Administration

  • Recommended dosage: Amlodipine and Atenolol 5/25 mg tablet once daily
  • Dosage may be increased as advised by physicians
  • Dosage should be individualized

Interaction

  • Disopyramide: Atenolol reduces the clearance of disopyramide by 20%
  • Ampicillin at high doses may reduce Atenolol levels
  • Oral antidiabetics and insulin: Beta-blockers may decrease tissue sensitivity to insulin and inhibit insulin secretion

Contraindications

  • Hypersensitivity to either component
  • Sinus bradycardia
  • Second and higher degrees of heart block
  • Cardiogenic shock
  • Hypotension
  • Congestive heart failure
  • Poor left ventricular function

  • Edema
  • Nausea
  • Drowsiness
  • Anxiety
  • Depression

Pregnancy & Lactation

  • Use during pregnancy only if benefit outweighs potential fetal risk
  • Not recommended for nursing mothers
  • Breast-feeding should be stopped if use is considered necessary

Precautions & Warnings

  • Use with caution in patients with airway obstruction
  • Can be used in patients with renal impairment
  • Caution in patients with severe liver damage
  • Patients should be warned against stopping the drug suddenly

Overdose Effects

  • Hypotension
  • Congestive cardiac failure in overdosage cases
  • Unabsorbed drugs may be removed by gastric lavage or charcoal administration
  • Symptomatic treatment suggested
