Indications
- Treatment of hypertension
- Lowering blood pressure
- Initial therapy for patients likely to need multiple antihypertensive agents

Pharmacology
- Amlodipine as a dihydropyridine calcium channel blocker
- Amlodipine's effect on vascular vs cardiac muscle cells
- Amlodipine as a peripheral arterial vasodilator
- Olmesartan Medoxomil's blocking of vasoconstrictor and aldosterone-secreting effects of angiotensin II
Dosage & Administration
- Substitution with individually titrated components
- Initiate therapy with 5/20 mg once daily for 1 to 2 weeks
- Titration up to a maximum of 10/40 mg once daily
- Recommended starting dose of Amlodipine for elderly patients
Interaction
- Attenuation of antihypertensive effect by NSAIDs including selective COX-2 inhibitors
- Close monitoring of blood pressure, renal function, and electrolytes
Contraindications
- Co-administration with Aliskiren in patients with diabetes

Pregnancy & Lactation
- Pregnancy Category D
- Contraindicated in 2nd and 3rd trimester
- Unknown excretion in human milk
- Adverse effects on nursing infant
Precautions & Warnings
- Risk of hypotension in volume- or salt-depleted patients
- Risk of vasodilation in patients with severe aortic stenosis
- Increased frequency, duration, or severity of angina or acute MI in patients with severe obstructive coronary artery disease
Use in Special Populations
- Safety and effectiveness not established in pediatric patients
- No significant differences in safety or effectiveness in subjects 65 years or older
- No studies in patients with renal impairment
- Initial therapy not recommended in hepatically impaired patients
