Description
- Indications: Vitamin D3 deficiency, Prevention of rickets, osteomalacia, osteoporosis, pre-eclampsia during pregnancy, immunity boost
- Pharmacology: Calcitriol binding to Vitamin D receptors, fat-soluble, absorption in small intestine, metabolized to 25-hydroxy Vitamin D3 and 1,25-dihydroxy Vitamin D3, stored in tissues, elimination via faeces and urine
- Dosage & Administration:
- Adult dosage: 40000 IU once weekly for 7 weeks for deficiency, 1400-2000 IU/day for maintenance, 20000 IU every 4 weeks for prevention
- Children dosage: 20000 IU once every 2 weeks for 6 weeks for deficiency, 20000 IU every 6 weeks for prevention
- Different dosage forms: capsule, film-coated tablet, oroflash/chewable tablets, syrup, injection
- Interaction: Drugs causing Vitamin D3 deficiencies, such as Magnesium containing Antacids, Digoxin, Thiazide Diuretics, etc.
- Contraindications: Hypercalcaemia, hypersensitivity to Vitamin D3, evidence of Vitamin D3 toxicity
- Side Effects: Anorexia, lassitude, nausea, vomiting, diarrhea, constipation, weight loss, polyuria, sweating, headache, raised concentrations of calcium and phosphate in plasma and urine
- Pregnancy & Lactation: Safe use up to 4000 IU during pregnancy, excreted in breast milk, overdose in infants induced by nursing mothers not observed
- Precautions & Warnings: Plasma-calcium concentration monitoring, caution in renal impairment, liver or kidney disease, primary hyperthyroidism, lymphoma, tuberculosis, granulomatous disease
- Use in Special Populations: Safety & efficacy not established in children under 12 years
- Overdose Effects: Hypervitaminosis D
