Indications

  • Nausea and vomiting of pregnancy in women who do not respond to conservative management

Pharmacology

  • Antihistamine
  • Blocks Histamine (H1) receptor
  • Crosses blood brain barrier
  • High affinity for H1 receptors in the brain
  • Inhibits vestibular system & Muscarinic receptor
  • Affects vomiting center
  • Antiemetic properties of Pyridoxine Hydrochloride

Dosage & Administration

  • Initial dose: one tablet orally at bedtime
  • Maximum dose: two tablets per day
  • Take on an empty stomach with water
  • Swallow tablets whole
  • Do not crush, chew, or split tablets
  • Take daily and not on an as needed basis

Interaction

  • Contraindicated with monoamine oxidase inhibitors (MAOIs)
  • Not recommended with alcohol and other CNS depressants

Contraindications

  • Hypersensitivity to Doxylamine Succinate, Pyridoxine Hydrochloride, or any inactive ingredient
  • Use with MAO inhibitors

Side Effects

  • Somnolence or accidents with CNS depressants

  • Pregnancy category A
  • Excreted into breast milk
  • Caution during breastfeeding

Precautions & Warnings

  • May cause somnolence
  • Avoid activities requiring mental alertness
  • Caution in women with asthma, increased intraocular pressure, narrow angle glaucoma, stenosing peptic ulcer, pyloroduodenal obstruction, or urinary bladder-neck obstruction

Overdose Effects

  • Extended-release formulation
  • Signs and symptoms may not be apparent immediately
  • Symptoms may include restlessness, dry mouth, dilated pupils, sleepiness, vertigo, mental confusion, tachycardia
  • At toxic doses: seizures, rhabdomyolysis, acute renal failure, death
  • Treatment: gastric lavage, activated charcoal, whole bowel irrigation, symptomatic treatment
