Indications
- Adjunct to diet and exercise for type 2 diabetes mellitus
- Not for type 1 diabetes patients
Pharmacology
- DPP-4 inhibitor
- Increases and prolongs action of incretin hormones
- Regulates glucose homeostasis
Dosage & Administration
- Recommended dose of 50mg twice a day or 100mg once daily
- Can be taken with or without food
Interaction
- No meaningful alteration of pharmacokinetics of various drugs
- Slightly increases mean Digoxin concentration
Contraindications
- History of serious hypersensitivity reactions to Sitagliptin
Side Effects
- Common adverse reactions include headache, upper respiratory tract infection and nasopharyngitis
- Potential for hypoglycemia
Pregnancy & Lactation
- Pregnancy Category B
- Secreted in the milk of lactating rats
Precautions & Warnings
- Prompt discontinuation if pancreatitis suspected
- Dosage adjustment in renal insufficiency
- Use with caution in hypersensitivity reactions
Use in Special Populations
- Safety and effectiveness not established in pediatric patients
- Careful dose selection in elderly with decreased renal function
Overdose Effects
- Maximal mean increases in QTc observed at high doses
- Supportive measures recommended in case of overdose
