Indications
- Treatment of invasive aspergillosis
- Treatment of candidemia and disseminated candidiasis
- Treatment of esophageal candidiasis
- Treatment of serious infections caused by specific fungal species
- Treatment for patients intolerant or refractory to other therapy
Pharmacology
- Voriconazole binds and inhibits ergosterol synthesis
- Inhibits CYP450-dependent 14-alpha sterol demethylase
- Results in depletion of ergosterol in fungal cell membrane
Dosage & Administration
- Oral tablet and powder for suspension
- Different loading and maintenance dose regimens based on body weight for oral administration
- Specific loading and maintenance dose regimens for injection administration based on infection type
Interaction
- Adjust dosage with CYP3A4, CYP2C9 and CYP2C19 inhibitors and inducers
- Potential to increase concentrations and activity of drugs that are CYP3A4, CYP2C9 and CYP2C19 substrates
- Increase maintenance dosage with co-administration of specific drugs
Contraindications
- Known hypersensitivity to Voriconazole or any other components
- Co-administration with specific drugs due to risk of serious adverse reactions or loss of efficacy
Side Effects
- Common side effects include abdominal pain, anemia, blurred vision, headache, chest pain, nausea, and diarrhea
Pregnancy & Lactation
- No adequate and well-controlled studies in pregnant women
- Use during pregnancy only if the potential benefit justifies the potential risk to the fetus
Precautions & Warnings
- Long term exposure over 180 days requires careful assessment of benefit-risk balance
- Reported cases of squamous cell carcinoma of the skin with long-term treatment
