Pharmacology

Pregnancy & Lactation

  • Associated with increased risk of congenital abnormalities and perinatal mortality if uncontrolled during pregnancy
  • Excreted into human breast milk, limited data for breastfeeding

Precautions & Warnings

  • Substantially excreted by the kidney, risk of accumulation and lactic acidosis
  • Lowering of vitamin B12 levels
  • Increased risk of hypoglycemia in combination with insulin
  • Regular monitoring of renal function advised

Use in Special Populations

  • Dosage adjustment in elderly subjects based on renal function
  • No effect on growth and puberty detected during controlled clinical studies
  • Caution recommended for children aged between 10 and 12 years

Overdose Effects

  • Hypoglycemia not seen at high doses, but lactic acidosis a possibility
  • Lactic acidosis considered a medical emergency
  • Hemodialysis as the most effective method to remove lactate and Metformin
