Pharmacology

Side Effects

Pregnancy & Lactation
- Associated with increased risk of congenital abnormalities and perinatal mortality if uncontrolled during pregnancy
- Excreted into human breast milk, limited data for breastfeeding
Precautions & Warnings
- Substantially excreted by the kidney, risk of accumulation and lactic acidosis
- Lowering of vitamin B12 levels
- Increased risk of hypoglycemia in combination with insulin
- Regular monitoring of renal function advised
Use in Special Populations
- Dosage adjustment in elderly subjects based on renal function
- No effect on growth and puberty detected during controlled clinical studies
- Caution recommended for children aged between 10 and 12 years
Overdose Effects
- Hypoglycemia not seen at high doses, but lactic acidosis a possibility
- Lactic acidosis considered a medical emergency
- Hemodialysis as the most effective method to remove lactate and Metformin
