Pharmacology
- Two well-known compounds: flupentixol and melitracen
- Flupentixol - a neuroleptic with anxiolytic and antidepressant properties
- Melitracen - a bipolar thymoleptic with activating properties
- Combination renders a preparation with antidepressant, anxiolytic, and activating properties
- Maximal serum concentration reached in about 4 hours after oral administration
- Biological half-life of flupentixol about 35 hours
- Biological half-life of melitracen about 19 hours
Dosage & Administration
- Adults: Usually 2 tablets orally daily in the morning and noon
- Severe cases may require an increased morning dose
- Elderly patients: 1 tablet in the morning
- Maintenance dose: Usually 1 tablet orally in the morning
Interaction
- Enhances the response to alcohol, barbiturates, and other CNS depressants
- Simultaneous administration of MAO-inhibitors may cause hypertensive crises
- Reduces the antihypertensive effect of guanethidine and similar acting compounds
- Enhances the effects of adrenaline and noradrenaline
Contraindications
- Immediate recovery phase after myocardial infarction
- Defects in bundle-branch conduction
- Untreated narrow-angle glaucoma
- Acute alcohol, barbiturate, and opiate intoxications
- Not recommended for excitable or overactive patients
- Patients who have received an MAO-inhibitor within two weeks
Side Effects
- Rare at recommended doses
- Transient restlessness
- Insomnia
Pregnancy & Lactation
- Preferably not to be given during pregnancy and lactation
Precautions & Warnings
- Gradual withdrawal of previous treatment with sedative tranquillizers
Overdose Effects
- Symptoms dominated by intoxications by melitracen, especially of anticholinergic nature
- Extrapyramidal symptoms due to flupentixol may occur
- Symptomatic and supportive measures required
- Gastric lavage and activated charcoal administration recommended
- Measures aimed at supporting the respiratory and cardiovascular systems
- Use of epinephrine (adrenaline) must be avoided
- Treatment for convulsions and extrapyramidal symptoms
