Indications
- Newly Diagnosed Glioblastoma Multiforme
- Refractory Anaplastic Astrocytoma
Pharmacology
Temozolomide undergoes rapid nonenzymatic conversion at physiologic pH to the reactive compound Methyl Triazen Imidazole Carboxamide (MTIC).
Dosage
- 75 mg/m^2 for 42 days concomitant with radiotherapy
- Initial maintenance dose of 150 mg/m^2 once daily for 6 cycles
Administration
Should be taken on an empty stomach at least 1 hr before meals.
Interaction
Administration of valproic acid decreases oral clearance of Temozar by about 5%.

- Hypersensitivity to any components
- Hypersensitivity to DTIC

Overdose Effects
Dose-limiting toxicity was hematologic. Adverse reactions may include pancytopenia, pyrexia, multi-organ failure, and death.
