Indications
- Prevention of rickets

Pharmacology
- Active form: Calcitriol
- Binding to Vitamin D receptors (VDRs)
- Fat soluble with a half-life of about 50 days
- Absorbed in the small intestine and transported to the liver
Dosage & Administration
- Adults dosage: 40000 IU once weekly for 7 weeks for Vitamin D3 deficiency
- Children dosage: 20000 IU once every 2 weeks for 6 weeks for Vitamin D3 deficiency
- Different dosage forms: capsule, film-coated tablet, oroflash, chewable tablets, syrup, injection
Interaction
- Many drugs cause Vitamin D3 deficiencies
- Some drugs interfere with the absorption and metabolism of Vitamin D3
Contraindications
- Contraindicated in diseases associated with hypercalcemia
- Contraindicated in patients with known hypersensitivity to Vitamin D3
Side Effects
- Symptoms include anorexia, nausea, constipation, headache, raised concentrations of calcium and phosphate in plasma and urine
Pregnancy & Lactation
- Safe use of doses up to 4000 IU during pregnancy
- Vitamin D3 and its metabolites are excreted in breast milk
Precautions & Warnings
- Plasma-calcium concentration should be monitored
- Consult a health care practitioner before supplementing with Vitamin D3 in certain conditions
Use in Special Populations
- Safety & efficacy in children under 12 years not established
